北京大學(xué)預(yù)防醫(yī)學(xué)學(xué)士、碩士、博士畢業(yè)(1989、1999、2006)，曾多次作為訪問學(xué)者赴美國Vanderbilt大學(xué)、Mayo Clinical醫(yī)學(xué)中心、德國慕尼黑工業(yè)大學(xué)進(jìn)行學(xué)術(shù)交流。現(xiàn)任北京大學(xué)臨床腫瘤學(xué)院副院長、流行病學(xué)研究室主任。中國抗癌協(xié)會(huì)腫瘤流行病學(xué)專業(yè)委員會(huì)副主任委員和青年委員會(huì)主任委員、國際胃癌協(xié)會(huì)委員、中國抗癌協(xié)會(huì)腫瘤標(biāo)志委員會(huì)委員、《中華流行病學(xué)雜志》編委等。先后承擔(dān)了多項(xiàng)國家自然科學(xué)基金和北京市自然科學(xué)基金、北京市科委重大項(xiàng)目子項(xiàng)目;作為主要人員參加了國家“973”、"863”項(xiàng)目、科技部“國際合作”重大專項(xiàng)及與美國NCI、德國慕尼黑工業(yè)大學(xué)合作項(xiàng)目等。在《Gut》、《Gastroenterology》、《Human Mutation》、《Cancer Research》等權(quán)威雜志上發(fā)表研究論文70余篇。獲得北京市科技進(jìn)步獎(jiǎng)及中華醫(yī)學(xué)科技獎(jiǎng)3項(xiàng)。

主要研究領(lǐng)域

目前主要從事胃癌分子流行病學(xué)研究，包括基因多態(tài)性與胃癌易感性、胃癌相關(guān)標(biāo)志物的篩選及干預(yù)相關(guān)分子機(jī)制研究。

代表性研究成果

1、發(fā)現(xiàn)了一組炎癥相關(guān)分子多態(tài)、表達(dá)與胃癌及癌前病變進(jìn)展密切相關(guān)

首次在大樣本人群中證明炎癥通路重要分子COX-2表達(dá)與重度胃癌癌前病變的發(fā)病風(fēng)險(xiǎn)密切相關(guān)，COX-2表達(dá)陽性者發(fā)生異型增生的風(fēng)險(xiǎn)增高5.35倍(OR=5.35， 95%CI 2.64-10.8)。進(jìn)一步分析發(fā)現(xiàn)COX-2 -1195G/A多態(tài)與COX-2表達(dá)及胃癌發(fā)病風(fēng)險(xiǎn)相關(guān)，并與幽門螺桿菌(H. pylori)感染存在明顯交互作用。該研究成果發(fā)表在Gastroenterology (IF=18.19)，同期雜志發(fā)表了述評(píng)，評(píng)價(jià)該研究揭示了COX-2、炎癥反應(yīng)在胃癌發(fā)生過程中的作用。另外，首次在大樣本人群中證明IL-8-251等炎癥反應(yīng)因子多態(tài)與胃癌發(fā)病風(fēng)險(xiǎn)密切相關(guān)(Carcinogenesis 2005, IF=5.108),與H. pylori感染存在協(xié)同作用。該研究被Clinical Microbiology Reviews等雜志引用149次。

2、胃癌相關(guān)標(biāo)志物的篩選及干預(yù)相關(guān)分子機(jī)制研究

利用山東臨朐胃癌高發(fā)現(xiàn)場長期隨訪隊(duì)列，篩選胃癌相關(guān)遺傳易感、表觀遺傳及血清蛋白標(biāo)志物，并通過檢測不同胃黏膜病變患者血清中H. pylori特異性抗體水平，預(yù)測H. pylori感染高危人群。研究發(fā)現(xiàn)胃黏膜病變進(jìn)展與H. pylori陽性抗體數(shù)目正相關(guān)、TLR1/10多態(tài)與H. pylori易感性相關(guān)、一組miRNA(miR-221, miR-376c, miR-744)表達(dá)水平及IGFII，N33甲基化水平在胃癌發(fā)生5年前顯著升高。通過檢測COX-2抑制劑干預(yù)前后胃黏膜細(xì)胞增殖/凋亡水平、COX-2甲基化及表達(dá)水平，發(fā)現(xiàn)干預(yù)效果與相關(guān)分子水平改變一致，證明H. pylori感染導(dǎo)致的炎癥反應(yīng)在胃癌發(fā)生過程中起重要作用，COX-2是預(yù)防胃癌有效的干預(yù)靶點(diǎn)。
