美國University of Texas MD Anderson Cancer Center和美國 University of Virginia Cancer Center博士后(1994-1998),瑞典Karolinska Institutet, Department of Laboratory Medicine, Division of Pathology助理教授、副教授(1998-2008),北京大學(xué)教授(2008-)?,F(xiàn)任北京大學(xué)人體解剖與組織胚胎學(xué)系主任、腫瘤研究中心副主任、基礎(chǔ)醫(yī)學(xué)院分子細(xì)胞生物學(xué)與腫瘤生物學(xué)實(shí)驗(yàn)室主任。作為課題組長,獲瑞典卡羅琳斯卡醫(yī)科大學(xué)、瑞典醫(yī)學(xué)會(huì)、瑞典癌癥研究基金會(huì)和瑞典研究委員會(huì)和北京大學(xué)985 II期工程等的多項(xiàng)科學(xué)基金資助。目前擔(dān)任國家科技部973課題負(fù)責(zé)人、國家自然科學(xué)基金面上及重點(diǎn)項(xiàng)目的負(fù)責(zé)人、教育部博士點(diǎn)基金和北京市自然科學(xué)基金重大項(xiàng)目負(fù)責(zé)人。1992年獲軍隊(duì)科學(xué)技術(shù)進(jìn)步一等獎(jiǎng)。1993年獲軍事醫(yī)學(xué)科學(xué)院優(yōu)秀青年科技工作者稱號(hào)。2006年獲瑞典卡羅琳斯卡醫(yī)科大學(xué)癌癥研究年度一等獎(jiǎng)。

主要研究領(lǐng)域
腫瘤侵襲轉(zhuǎn)移的分子細(xì)胞生物學(xué)機(jī)制。主要研究興趣在跨膜受體整合素胞內(nèi)結(jié)構(gòu)域相互作用蛋白所介導(dǎo)的腫瘤增殖、侵襲、轉(zhuǎn)移和網(wǎng)絡(luò)調(diào)控的分子細(xì)胞生物學(xué)機(jī)制,腫瘤細(xì)胞重編程產(chǎn)生腫瘤干細(xì)胞及其在腫瘤復(fù)發(fā)和轉(zhuǎn)移中的作用機(jī)制。
代表性研究成果
1. 發(fā)現(xiàn)整合素相關(guān)蛋白Kindlin-2在前列腺癌細(xì)胞中調(diào)控一線抗前列腺癌藥物順鉑的敏感性。首先揭示敲低整合素相關(guān)蛋白Kindlin-2會(huì)提高順鉑對前列腺癌細(xì)胞的殺傷能力。該研究結(jié)果2010年發(fā)表在《Cancer Lett》上,并被推薦為藥物領(lǐng)域Faculty 1000必讀論文。
2. 發(fā)現(xiàn)整合素相關(guān)蛋白Kindlin-2可以在細(xì)胞核內(nèi)與Wnt信號(hào)通路的重要調(diào)控分子β-catenin及Wnt信號(hào)的重要靶基因TCF4形成三分子復(fù)合物,增強(qiáng)Wnt信號(hào)通路的激活。首先揭示出Kindlin-2在細(xì)胞核內(nèi)的生物學(xué)功能及調(diào)控EMT的復(fù)雜機(jī)制。該研究結(jié)果2012年發(fā)表在《EMBO Rep》上,并被推薦為細(xì)胞生物學(xué)領(lǐng)域Faculty 1000必讀論文。
3. 發(fā)現(xiàn)整合素相關(guān)蛋白Kindlin-2在腎上皮細(xì)胞中與TGF-β I型受體及Smad3相互作用,調(diào)控EMT及腎臟的纖維化。該研究結(jié)果2013年發(fā)表在《J Am Soc Nephrol》上,雜志還撰寫了專題評述。
4. 發(fā)現(xiàn)組蛋白甲基轉(zhuǎn)移酶EZH2可以被乙酰轉(zhuǎn)移酶PCAF乙酰化;EZH2還可以被去乙?;窼IRT1去乙?;?乙酰化的EZH2可以增加自身的穩(wěn)定性,并在肺腺癌的預(yù)后中起作用。該研究結(jié)果2015年發(fā)表在《Nucleic Acids Research》上。
